{"id":1119,"date":"2012-06-04T10:04:21","date_gmt":"2012-06-04T09:04:21","guid":{"rendered":"https:\/\/blogs.bmj.com\/heart-journalscan\/?p=1119"},"modified":"2015-11-12T15:58:04","modified_gmt":"2015-11-12T14:58:04","slug":"microrna-214-attenuates-ischaemia-reperfusion-injury","status":"publish","type":"post","link":"https:\/\/blogs.bmj.com\/heart\/2012\/06\/04\/microrna-214-attenuates-ischaemia-reperfusion-injury\/","title":{"rendered":"MicroRNA-214 attenuates ischaemia-reperfusion injury"},"content":{"rendered":"<p style=\"text-align: justify\">Ischaemia reperfusion injury (IR) is characterised by\u00a0 abnormal increases in intracellular calcium during myocardial reperfusion that lead to cardiomyocyte death.\u00a0 In addition, IR is accompanied by an increase in the expression of microRNAs (miRNAs) such as miR-214, which has previously been identified as a sensitive marker of cardiac stress.<!--more--><\/p>\n<p style=\"text-align: justify\">In this study, Aurora et al. found that genetic deletion of miR-214 in mice was associated with an increase in cardiomyocyte apoptosis, loss of cardiac contractility, and excessive fibrosis following IR injury.\u00a0 Subsequent work demonstrated that the cardioprotective role of miR-214 was due to its repression the mRNA encoding a key regulator of calcium influx, the Ncx1 sodium\/calcium exchanger, and also to repression of several downstreatm effectors of calcium signalling that mediate cell death.<\/p>\n<p style=\"text-align: justify\"><strong>Conclusions:<\/strong><\/p>\n<p style=\"text-align: justify\">This paper reveals a key role for the microRNA miR-214 in the response to ischaemia-reperfusion injury through its influence on calcium homeostasis.\u00a0 Therefore, increasing levels of miR-214 to attentuate calcium overload &#8211; and therefore cell death &#8211; could be a new therapeutic strategy for limiting ischaemia-reperfusion injury.<\/p>\n<ul>\n<li>Aurora AB, Mahmoud AI, Luo X et al.\u00a0 MicroRNA-214 protects the mouse heart from ischemic injury by controlling Ca<sup>2+<\/sup> overload and cell death.\u00a0 <em>J Clin Invest<\/em> 2012;122:1222-1232.<\/li>\n<\/ul>\n<p><!--TrendMD v2.4.8--><\/p>\n","protected":false},"excerpt":{"rendered":"<p>Ischaemia reperfusion injury (IR) is characterised by\u00a0 abnormal increases in intracellular calcium during myocardial reperfusion that lead to cardiomyocyte death.\u00a0 In addition, IR is accompanied by an increase in the expression of microRNAs (miRNAs) such as miR-214, which has previously been identified as a sensitive marker of cardiac stress.
