Our <a href=\"https:\/\/fdaaa.trialstracker.net\/\" target=\"_blank\" rel=\"noopener\">FDAAA TrialsTracker<\/a> shows all individual trials that <a href=\"https:\/\/www.biorxiv.org\/content\/early\/2018\/03\/12\/266452.full.pdf\" target=\"_blank\" rel=\"noopener\">breach<\/a> this legal requirement. Once a week, we write about one unreported clinical trial: you can read more background <a href=\"https:\/\/blogs.bmj.com\/bmj\/2018\/03\/29\/it-is-time-to-fix-the-problem-of-unreported-clinical-trials\/\" target=\"_blank\" rel=\"noopener\">here<\/a>.<b><\/b><\/p>\n<p dir=\"ltr\"><strong>Trial NCT00450463<\/strong><\/p>\n<p dir=\"ltr\">This week\u2019s unreported trial is titled: \u201cA Randomized Phase II Trial Combining Vaccine Therapy With PROSTVAC\/TRICOM and Flutamide vs. Flutamide Alone in Men With Androgen Insensitive, Non-Metastatic (D0.5) Prostate Cancer\u201d (<a href=\"https:\/\/clinicaltrials.gov\/ct2\/show\/NCT00450463\" target=\"_blank\" rel=\"noopener\">NCT00450463<\/a>). The trial enrolled 53 participants with non-metastatic androgen insensitive prostate cancer. It was designed as a crossover study. The experimental intervention was flutamide in addition to a priming vaccine (PROSTVAC-V\/TRICOM), a booster vaccine (PROSTVAC-F\/TRICOM), and a chemical that boosts immune activity (GM-CSF); the active comparator was flutamide only. The primary outcome was time to treatment failure over 4-5 years of follow-up. The secondary outcomes were: time to development of metastatic disease; toxicity; response rate; PSA; and immune responses and Immunologic effects.<b><\/b><\/p>\n<p dir=\"ltr\"><strong>Clinical Discussion<\/strong><\/p>\n<p dir=\"ltr\"><a href=\"https:\/\/www.europeanurology.com\/article\/S0302-2838(16)30251-2\/fulltext\/global-incidence-and-mortality-for-prostate-cancer-analysis-of-temporal-patterns-and-trends-in-36-countries\" target=\"_blank\" rel=\"noopener\">Recent estimates<\/a> put the global age-standardized rate of prostate cancer at 30.6 per 100,000 with incidence on the rise throughout much of the world. It is the <a href=\"https:\/\/seer.cancer.gov\/statfacts\/html\/prost.html\" target=\"_blank\" rel=\"noopener\">third most common<\/a> form of cancer in the United States and the <a href=\"http:\/\/www.cancerresearchuk.org\/health-professional\/cancer-statistics\/incidence\/common-cancers-compared#heading-Zero\" target=\"_blank\" rel=\"noopener\">second most common<\/a>, along with lung cancer, in the United Kingdom. Androgen insensitive prostate cancer is difficult to treat. Vaccines for prostate cancer have <a href=\"https:\/\/www.tandfonline.com\/doi\/full\/10.1586\/erv.13.27\" target=\"_blank\" rel=\"noopener\">shown some promise<\/a> and one therapy is already FDA approved for use in castration refractory prostate cancer. The PROSTVAC\/TRICOM vaccine has shown particular promise in another <a href=\"https:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2834462\/\" target=\"_blank\" rel=\"noopener\">phase 2 trial<\/a> and is the subject of a recently completed <a href=\"https:\/\/clinicaltrials.gov\/ct2\/show\/results\/NCT01322490\" target=\"_blank\" rel=\"noopener\">phase 3 trial<\/a>. Knowing the benefits of this potential treatment for an extremely common form of cancer, compared to a standard of care such as flutamide, is essential research for cancer patients and doctors alike.<b><\/b><\/p>\n<p dir=\"ltr\"><strong>Conclusion<\/strong><\/p>\n<p dir=\"ltr\">This unreported trial was sponsored by the National Cancer Institute in collaboration with the National Institutes of Health Clinical Center. The study PI is Dr. Ravi A Madan of the National Cancer Institute. As of 13 June 2018, this trial is 47 days overdue to report.
